目的 将平行平板流动装置用于体外药物试验研究 ,为心血管药物的筛选和测试提供新的实验方法和手段。方法 用放射免疫法测定内皮素 (ET- 1)和 6 -酮 -前列腺素 F1α(6 - keto- PGF1α)的分泌速率和累积分泌量 ,观察复方丹参注射液、丹皮酚和阿司匹林等心血管药物在剪切流场中对内皮细胞分泌 ET- 1和前列腺素 (PGI2 )功能的干预作用。结果 复方丹参注射液和丹皮酚对 ET- 1、PGI2 的分泌功能均有促进作用 ,而阿司匹林则无影响。结论 平行平板流动装置条件易控制 ,观察和测定方便 ,不失为一种研究心血管药物对内皮细胞功能影响的较好方法。实验结果有利于对丹参和丹皮酚的血管作用有新的认识
OBJECTIVE: To use parallel plate flow devices for in vitro drug testing and to provide new experimental methods and tools for the screening and testing of cardiovascular drugs. Methods The secretion rate and cumulative secretion of ET - 1 and 6 - keto - prostaglandin F1α (6 - keto - PGF1α) were measured by radioimmunoassay. The effects of Fufang Danshen injection, paeonol and aspirin on cardiovascular Interventional effect of drugs on endothelial cell secretion of ET-1 and prostaglandin (PGI2) in shear flow field. Results Salvia miltiorrhiza injection and paeonol promoted the secretion of ET-1 and PGI2, while aspirin had no effect. Conclusion The parallel plate flow device is easy to control and convenient to observe and measure. It is a good method to study the effect of cardiovascular drugs on endothelial cell function. The experimental results are conducive to the new understanding of the blood vessels of Salvia miltiorrhiza and paeonol
