目的:建立适合混合蛋白质分离分析的高效毛细管电泳方法。方法:采用高效毛细管电泳中自由区带电泳分离模式,以标准蛋白质细胞色素C与溶菌酶为分析对象,对影响电泳分离的因素进行了考察。结果与结论:通过减少蛋白质管壁吸附、优选缓冲液体系及定性定量信息校正,使细胞色素C与溶菌酶获得了理想的分离,同时成功地应用于磷脂酶A2的优化分离与纯度分析,为蛋白质类生化药物的质量控制提供了简便、快速、有效的检测手段
Objective: To establish a high performance capillary electrophoresis method suitable for the separation and analysis of mixed proteins. Methods: High performance capillary electrophoresis with free zone electrophoresis separation mode, the standard protein cytochrome C and lysozyme for the analysis of the factors that affect electrophoretic separation were investigated. RESULTS AND CONCLUSION: Cytochrome C and lysozyme were ideally separated by reducing protein tube wall adsorption, optimizing buffer system and qualitative and quantitative information, and were successfully applied to the optimization and purity analysis of phospholipase A2 Quality control of protein biochemical drugs provides a simple, rapid and effective means of detection
