以Cd基因5’cDNA片段为探针,经DNA印迹杂交法对北京地区60例正常无关汉族个体进行了C4基因限制性片段长度多态性(RFLP)分析。以限制性内切酶HindⅢ消化基因组DNA时检测到5种限制性片段长度变型(RFLV),它们的组成及出现频率为,A:32-25-15 kb(0.433)、B:32-15kb(0.383)、C:25-15 kb(0.117)、D:32-15-8.5 kb(0.05)及E:25-15-8.5(0.017)。此外,还对表现C4A基因缺失的部分样本及数例无C4A基因缺失样本以限制性内切酶BamHⅠ消化其基因组DNA,检出3种RFLV,A:4.5-3.3kb、B:4.5kb及C:3.3kb。
The restriction endonuclease fragment length polymorphism (RFLP) analysis of C4 gene in 60 normal unrelated Han individuals in Beijing area was performed by Southern blotting with cDNA of Cd gene as a probe. Five restriction fragment length variants (RFLVs) were detected upon digestion of genomic DNA with the restriction endonuclease HindIII and their composition and frequency were: A: 32-25-15 kb (0.433), B: 32-15 kb 0.383), C: 25-15 kb (0.117), D: 32-15-8.5 kb (0.05) and E: 25-15-8.5 (0.017). In addition, three RFLVs, A: 4.5-3.3 kb, B: 4.5 kb and C (C) were also detected for partial samples showing deletion of the C4A gene and several cases of absence of the C4A gene from the genomic DNA digested with the restriction enzyme BamHI : 3.3kb.
