目的 :探讨内毒素、γ 干扰素对鼠巨噬细胞胆固醇清除受体 β类I型 (SR β1 )表达及功能的影响。 方法 :采用Northernblot及蛋白质免疫印迹法测定经内毒素和干扰素处理后的巨噬细胞SR β1受体在mRNA转录及蛋白翻译水平的表达。结果 :用内毒素处理巨噬细胞 8h可使SR β1受体在mRNA转录及蛋白翻译水平均下调 ( 5 4%和5 3 % ) ,而干扰素则可使之下调 5 7%和 5 4% ,并呈现浓度依赖性和时间依赖性。结论 :( 1 )内毒素、干扰素在mRNA和蛋白双重水平下调SR β1的表达 ,明显降低SR β1调节胆固醇代谢的作用。 ( 2 )内毒素通过降低SR β1的表达及功能 ,在动脉粥样硬化过程中起着不可忽视的作用。
Objective: To investigate the effects of endotoxin and interferon gamma on the expression and function of cholesterol-releasing receptor β-type I (SRβ1) in murine macrophages. Methods: Northern blotting and Western blotting were used to determine the mRNA and protein translation level of SRβ1 in endotoxin and interferon treated macrophages. Results: Treatment of macrophages with endotoxin for 8 h reduced the level of mRNA and protein translation of SRβ1 (54% and 53%), while interferon reduced the levels of SRβ1 by 57% and 54% , And showed concentration-dependent and time-dependent. Conclusion: (1) Endotoxin and interferon downregulate the expression of SRβ1 at both mRNA and protein levels, significantly reducing the effect of SRβ1 on cholesterol metabolism. (2) Endotoxin plays an important role in atherosclerosis by reducing the expression and function of SRβ1.
