目的探索成年糖尿病大鼠心室肌细胞成功分离的方法。方法取体质量150~200 g的Sprague-Daw ley大鼠20只,单次腹腔注射链脲佐菌素(STZ)建立DM大鼠模型,3周后取大鼠心脏行主动脉逆行灌流分离心室肌细胞;并在倒置显微镜下观察分离的单个心室肌细胞状态。结果建模成功率达90%,糖尿病大鼠的血糖水平稳定维持在20 mmol/L以上;分离的单个心室肌细胞存活率达70%~90%,且纹理清晰状态好。结论该糖尿病心室肌细胞的分离方法简单有效。
Objective To explore the method of successful isolation of ventricular myocytes from adult diabetic rats. Methods Twenty Sprague-Dawley rats weighing 150-200 g were given a single intraperitoneal injection of streptozotocin (STZ) to establish a rat model of DM. Three weeks later, aortic retrograde perfusion was used to separate the ventricles Muscle cells; and observe the status of isolated single ventricular myocytes under an inverted microscope. Results The success rate of modeling was 90%. The blood glucose level of diabetic rats remained stable at 20 mmol / L or more. The isolated single ventricular myocytes survival rate was 70% ~ 90% and the texture was clear. Conclusion The isolation of diabetic ventricular myocytes is simple and effective.
