目的 了解SIEA 2 6 2 8ku抗原分子的结构和功能以及生物学特性。 方法 采用梯度聚丙烯酰胺凝胶制备电泳和高效液相色谱方法 ,分离纯化SIEA2 6 2 8ku分子 ,并对其进行紫外吸收光谱分析鉴定。 结果 通过紫外吸收光谱分析表明 ,SIEA2 6 2 8ku抗原组分的紫外吸收光谱共有 6个特征性蛋白吸收峰 ,分别在 2 5 8、2 60、2 64 .5、2 68、2 69.5和 2 72nm ,部分吸收峰符合苯丙氨酸的吸收光谱。 结论 高效液相色谱技术结合紫外吸收光谱分析 ,可为日本血吸虫SIEA 2 6 2 8ku抗原组分结构和功能的进一步研究以及生物学特性分析提供重要信息。
Objective To understand the structure, function and biological characteristics of SIEA 2 6 2 8 ku antigen. METHODS: Gradient polyacrylamide gel electrophoresis and high performance liquid chromatography (HPLC) were used to separate and purify SIEA2 6 2 8ku molecules, and their UV-Vis absorption spectra were identified. Results The results of ultraviolet absorption spectroscopy showed that the UV absorption spectra of the SIEA2 6 2 8 ku antigen component had 6 characteristic protein peaks at 25 8,2 60,2 64.5,2 68,2 69.5 and 72 72 nm , Part of the absorption peak in line with phenylalanine absorption spectrum. Conclusion High performance liquid chromatography coupled with ultraviolet absorption spectroscopy can provide important information for the further study of the structure and function of SIEA 2 6 2 8 ku antigen components and the biological characteristics analysis of Schistosoma japonicum.
