目的探讨低剂量硝酸镧对糖尿病大鼠脂肪组织中过氧化物酶体增殖物激活受体-γ(peroxisome proliferatoractivated receptor-gamma,PPAR-γ)和葡萄糖转运蛋白4(glucose transporter protein 4,GLUT4)蛋白表达水平的影响。方法将40只健康清洁级Wistar雄性大鼠随机分为3组,分别为对照组(10只)、糖尿病组(18只)和硝酸镧组(12只)。糖尿病组和硝酸镧组腹腔注射50 mg/kg链脲佐菌素制备糖尿病模型;硝酸镧组采用灌胃方式染毒0.2 mg/kg硝酸镧,对照组及糖尿病组均每日给予等剂量的生理盐水,每天1次,连续染毒1个月。采用免疫组织化学方法检测大鼠脂肪组织PPAR-γ和GLUT4的蛋白表达水平,并观察脂肪组织的病理改变。结果与对照组比较,糖尿病组大鼠脂肪组织PPAR-γ和GLUT4的蛋白表达水平均降低,差异有统计学意义(P<0.01);而硝酸镧组大鼠脂肪组织PPAR-γ和GLUT4的蛋白表达水平未见明显变化。与糖尿病组比较,硝酸镧组大鼠脂肪组织PPAR-γ和GLUT4的蛋白表达水平升高,差异有统计学意义(P<0.01)。结论口服低剂量硝酸镧(0.2 mg/kg)可通过促进糖尿病大鼠脂肪组织中PPAR-γ和GLUT4蛋白的表达,从而调节大鼠脂肪组织的代谢进而起到保护作用。
Objective To investigate the effect of low dosage of lanthanum nitrate on the expression of peroxisome proliferator-activated receptor-gamma (PPAR-γ) and glucose transporter protein 4 (GLUT4) in adipose tissue of diabetic rats Effect of expression level. Methods Forty healthy male Wistar rats were randomly divided into three groups: control group (n = 10), diabetes group (n = 18) and lanthanum nitrate group (n = 12). Diabetic group and lanthanum nitrate group were injected intraperitoneally with streptozotocin 50 mg / kg to prepare diabetic model. The lanthanum nitrate group was treated with 0.2 mg / kg lanthanum nitrate by intragastric administration, while the control group and diabetic group were given equal doses of physiology Saline, 1 day, continuous exposure to 1 month. Immunohistochemistry was used to detect the protein expression of PPAR-γ and GLUT4 in adipose tissue of rats, and the pathological changes of adipose tissue were observed. Results Compared with the control group, the protein expression levels of PPAR-γ and GLUT4 in adipose tissue of diabetic rats decreased significantly (P <0.01), while the protein expressions of PPAR-γ and GLUT4 in adipose tissue of rats in Lanthanum nitrate group No significant changes in expression levels. Compared with the diabetic group, the protein expression of PPAR-γ and GLUT4 in the adipose tissue of the Lanthanum nitrate group increased significantly, with statistical significance (P <0.01). Conclusion Oral administration of low dose of lanthanum nitrate (0.2 mg / kg) can protect the metabolism of adipose tissue in rats by promoting the expression of PPAR-γ and GLUT4 protein in adipose tissue of diabetic rats.
