目的研究硫酸锌(ZnS)和葡萄糖酸锌(ZnG)在大鼠体内的动态组织分布,并获得药代动力学参数。方法大鼠分别灌胃4 mg/kg Zn(ZnS和ZnG)后1、4、8、12、24、48、72和120 h处死动物,并采集心、肝、脾、肺、肾、胰、脑、睾丸、骨骼肌、股骨组织样品。大鼠组织样品用硝酸与高氯酸混合酸消化处理,原子吸收分光光度法测定组织中锌浓度,非房室模型计算ZnS和ZnG的药代动力学参数。结果大鼠灌胃ZnS和ZnG在各组织中的末端消除半衰期较长,平均滞留时间约为60 h。除胰外,ZnG在各组织中的C_(max)均高于ZnS。结论 ZnS和ZnG广泛分布于大鼠各组织中,两者均在股骨中浓度最高,其后依次为肝、胰、睾丸、肾、脾、肺、心。
Objective To study the dynamic tissue distribution of zinc sulfate (ZnS) and zinc gluconate (ZnG) in rats and to obtain pharmacokinetic parameters. Methods Rats were sacrificed at 1, 4, 8, 12, 24, 48, 72 and 120 h after 4 mg / kg Zn (ZnS and ZnG) Brain, testis, skeletal muscle, femur tissue samples. Rat tissue samples were treated with nitric acid and perchloric acid mixed acid digestion, atomic absorption spectrophotometry to determine the concentration of zinc in the tissue, non-compartmental model calculated ZnS and ZnG pharmacokinetic parameters. Results The end-eradication half-lives of ZnS and ZnG in rats were longer than those in other groups. The average residence time was about 60 h. Except pancreas, the C_ (max) of ZnG in all tissues was higher than that of ZnS. Conclusion ZnS and ZnG are widely distributed in various tissues of rats. Both of them have the highest concentration in the femur, followed by liver, pancreas, testis, kidney, spleen, lung and heart.
