目的:研究ADAM10蛋白表达与口腔鳞状细胞癌淋巴结转移的相关性,并探讨其促进肿瘤细胞转移的可能机制。方法:采用免疫组织化学方法检测ADAM10蛋白在58例口腔鳞状细胞癌中的表达情况,并分析其与肿瘤淋巴结转移发生的相关性;采用Transwell法检测高、低转移潜能的HN-12和HN-4口腔鳞状细胞癌细胞系的迁移能力,并通过实时定量PCR法和蛋白质免疫印迹实验分别检测2株细胞中ADAM10基因和蛋白的表达水平;采用RNA干扰技术,沉默高转移潜能细胞HN-12中的ADAM10的表达水平,观察其对细胞迁移能力的影响。采用SPSS 16.0软件包对实验数据进行统计学分析。结果:ADAM10蛋白的高表达与口腔鳞状细胞癌发生淋巴结转移相关;在高转移潜能细胞系HN-12细胞中,ADAM10基因及蛋白表达水平较低转移潜能细胞系HN-4显著增高(P<0.01);HN-12细胞中沉默表达ADAM10基因后,细胞迁移能力显著降低(P<0.01)。结论:ADAM10蛋白的高表达与口腔鳞状细胞癌淋巴结转移相关;ADAM10促进肿瘤细胞的迁移能力是其影响肿瘤转移的可能机制。ADAM10有望成为治疗口腔鳞状细胞癌转移的新靶点。
Objective: To investigate the correlation between ADAM10 protein expression and lymph node metastasis in oral squamous cell carcinoma and to explore its possible mechanism of promoting tumor cell metastasis. Methods: Immunohistochemical method was used to detect the expression of ADAM10 in 58 cases of oral squamous cell carcinoma and its correlation with tumor lymph node metastasis. The transwell method was used to detect the expression of HN-12 and HN -4 oral squamous cell carcinoma cell line migration ability, and by real-time quantitative PCR and Western blotting were detected in two cells ADAM10 gene and protein expression levels; using RNA interference technology, high metastatic potential cell HN- 12 in the expression of ADAM10 to observe its impact on cell migration. SPSS 16.0 software package was used to analyze the experimental data. Results: The high expression of ADAM10 protein was associated with lymph node metastasis in oral squamous cell carcinoma. In high metastatic potential cell line HN-12, the expression level of ADAM10 protein and HN-4 in metastatic potential cell line was significantly higher (P < 0.01). The silencing of ADAM10 expression in HN-12 cells was significantly decreased (P <0.01). Conclusion: The overexpression of ADAM10 protein is associated with lymph node metastasis in oral squamous cell carcinoma. ADAM10 promotes the migration of tumor cells as a possible mechanism of its effect on tumor metastasis. ADAM10 is expected to become a new target for the treatment of oral squamous cell carcinoma metastasis.
