目的 :对海马神经元癫样放电后的程序性死亡神经元作定性和定量检测 ,以探讨颞叶癫患者海马神经元丢失的机制。 方法 :首先制备海马神经元癫样放电模型 ,然后采用DNA梯度电泳、Tunel标记、荧光染色以及流式细胞技术对模型中的程序性死亡神经元作定性和定量检测。 结果 :发现神经元癫样放电后出现细胞程序性死亡 ,而且随着放电时间的延长 ,程序性死亡细胞递增。 结论 :反复癫样放电的兴奋性毒性导致神经元程序性死亡 ,颞叶癫患者海马神经元丢失与多种因素有关
OBJECTIVE: To qualitatively and quantitatively detect apoptotic neurons after epileptiform discharge in hippocampal neurons to explore the mechanism of neuronal loss in hippocampus of patients with temporal lobe epilepsy. Methods: Firstly, epileptiform discharge model of hippocampal neurons was prepared. DNA ladder electrophoresis, Tunel labeling, fluorescent staining and flow cytometry were used to detect the programmed death neurons in the model. Results: Neuronal apoptosis was found after epileptiform discharge, and as the discharge time prolonged, the number of apoptotic cells increased. CONCLUSIONS: Excitotoxicity induced by repeated epileptiform discharges leads to neuronal programmed death.
