目的:探讨特发性血小板减少性紫癜(ITP)患者骨髓血小板生成素(Tpo)和其受体c-mpl基因变化的意义。方法:用非放射性核素细胞原位杂交法检测tpo、c-mpl mRNA;细胞免疫化学法检测巨核细胞MPL。结果:ITP患者骨髓tpo、c-mpl mRNA,巨核细胞MPL表达增高。结论:c-mpl基因是在转录水平调控的;人体骨髓内对tpo及其受体的调节是以负反馈方式进行的;ITP患者骨髓局部tpo表达增高,病理情况下巨核细胞MPL增高可能与循环Tpo水平降低有关。
Objective: To investigate the significance of changes in bone marrow thrombopoietin (Tpo) and its receptor c-mpl gene in patients with idiopathic thrombocytopenic purpura (ITP). Methods: Tpo and c-mpl mRNA were detected by in situ hybridization with non-radioactive nuclei; MPL was detected by immunocytochemistry. RESULTS: The expression of tpo, c-mpl mRNA in bone marrow and MPL in megakaryocytes increased in patients with ITP. Conclusion: The c-mpl gene is regulated at the transcriptional level; the regulation of tpo and its receptors in the human bone marrow is performed by negative feedback; the expression of tpo in the bone marrow of patients with ITP is increased, and the increase of MPL in megakaryocytes may be associated with circulation in pathological conditions. The level of Tpo is reduced.
