目的:造血干细胞的增殖和分化,可认为是由造血微环境调控的。本实验目的在于在体外重建干细胞和造血环境的相互作用,并可见到辐射后造血干细胞发生和在体内同样的恢复和增殖。现报告如下: 方法:骨髓细胞在含有20%马血清的Fischer培养基中培养三周。形成贴壁底层后,在其上再加1×10~7骨髓有核细胞继续培养。培养液每周更换半量,取其细胞悬液计算CFUs和CFUc。在骨髓有核细胞双层培养24小时后进行照射,剂量率每分钟250R。此外,还观察了照射后贴壁细胞集落的辐射敏感性。结果:1)以贴壁细胞集落为指标测定了造血微环境的辐射敏感性,其D_0值约为100R。2)在本培养
Purpose: Proliferation and differentiation of hematopoietic stem cells can be considered as regulated by hematopoietic microenvironment. The purpose of this experiment is to reconstruct the interaction of stem cells and hematopoietic environment in vitro, and to see the occurrence and subsequent recovery and proliferation of hematopoietic stem cells after irradiation. Now report as follows: Methods: Bone marrow cells were cultured in Fischer’s medium containing 20% horse serum for three weeks. After the formation of adherent bottom, plus 1 × 10 ~ 7 bone marrow nucleated cells continue to train. The medium was changed weekly by half and the CFU and CFUc were calculated from the cell suspension. Irradiation was performed 24 hours after bone marrow nucleated cells were cultured at a rate of 250R per minute. In addition, the radiosensitivity of adherent cell colonies after irradiation was also observed. Results: 1) The radiosensitivity of hematopoietic microenvironment was determined by adherent cell colonies with D0 value of about 100R. 2) In this training
