目的:探讨环氧化酶-2(COX-2)与血管内皮生长因子(VEGF)在人翼状胬肉成纤维细胞(HPF)中的关系,以及美洛昔康(Meloxicam)抗HPF增殖的作用和机制。方法:采用免疫组化方法测定翼状胬肉组织中COX-2的表达。体外培养HPF,采用Western法,MTT法,研究在不同浓度的VEGF刺激下,HPF中COX-2蛋白的表达,以及不同浓度的美洛昔康对HPF增殖的影响。结果:翼状胬肉组织中COX-2呈阳性表达。HPF中VEGF浓度的增高可致COX-2蛋白的表达呈增强趋势。当美洛昔康的浓度在75~300μmol/L之间变化时,能明显抑制HPF的增殖,且其抑制强度呈剂量、时间依赖性。结论:HPF中VEGF的浓度升高能增强COX-2蛋白的表达,美洛昔康能够通过拮抗COX-2达到抑制HPF增殖的作用。
Objective: To investigate the relationship between cyclooxygenase-2 (COX-2) and vascular endothelial growth factor (VEGF) in human pterygium fibroblasts (HPF) and the effect of meloxicam on HPF proliferation And mechanism. Methods: Immunohistochemistry was used to determine the expression of COX-2 in pterygium. HPF was cultured in vitro. Western blot and MTT assay were used to investigate the expression of COX-2 in HPF and the effect of different concentrations of meloxicam on the proliferation of HPF. Results: COX-2 was positively expressed in pterygium. Increased VEGF concentration in HPF results in an increase in COX-2 protein expression. When the concentrations of meloxicam varied from 75μmol / L to 300μmol / L, the proliferation of HPF could be obviously inhibited, and the intensity of inhibition was dose-and time-dependent. CONCLUSION: The increase of VEGF concentration in HPF can enhance the expression of COX-2 protein. Meloxicam can inhibit the proliferation of HPF by antagonizing COX-2.
