目的分析2013年珠海市手足口病(Hand-foot-mouth disease,HFMD)患者中,肠道病毒71型(EV71)和柯萨奇病毒A组16型(Cox A16)的分子流行病学特征。方法采集珠海市2013年HFMD临床诊断病例粪便标本215份,选取其中Real time RT-PCR检测EV71病毒核酸阳性的18份和Cox A16病毒核酸阳性的8份标本,进行VP4区基因扩增,并进行核苷酸序列和遗传进化关系分析。结果 16份EV71扩增测序成功,与C4a基因亚型代表株具有最高的核苷酸序列同源性(95.2%~99.5%)和氨基酸同源性(91.3%~100%);5份Cox A16扩增测序成功,有2份与B2a亚型代表株具有最高的核苷酸序列同源性(98.6%~99.0%)和氨基酸同源性(97.1%~98.5%),3份与B2b亚型代表株具有最高的核苷酸序列同源性(96.6%~97.1%)和氨基酸同源性(100%)。结论 2013年珠海市流行的EV71属于C4a基因亚型,Cox A16为B2a和B2b两种基因亚型。
Objective To analyze the molecular epidemiological characteristics of EV71 and Cox A16 in Hand-foot-mouth disease (HFMD) patients in 2013 in Zhuhai. Methods A total of 215 stool specimens from HFMD in Zhuhai City in 2013 were collected. Among them, 18 were positive for EV71 virus and 8 were positive for Cox A16 virus by Real time RT-PCR. Analysis of Nucleotide Sequence and Genetic Evolution. Results Sixteen EV71 sequences were successfully sequenced and showed the highest nucleotide sequence homology (95.2% -99.5%) and amino acid homology (91.3% -100%) with C4a subtype representative strains. Five copies of Cox A16 The two strains showed the highest nucleotide sequence homology (98.6% -99.0%) and amino acid homology (97.1% -98.5%) with the representative strains of B2a subtype, and three copies of B2b subtype The representative strain has the highest nucleotide sequence homology (96.6% ~ 97.1%) and amino acid homology (100%). Conclusion The EV71 epidemic in Zhuhai in 2013 belongs to the C4a genotype, and Cox A16 is B2a and B2b genotypes.
