白薇煎对糖尿病神经病变大鼠脊神经致痛通道蛋白的影响

目的:观察中药复方白薇煎对糖尿病大鼠脊神经传导速度和脊神经致痛通道蛋白的影响。方法:大鼠随机选取10只为正常组,其余为模型组,采用链尿佐菌素(STZ)造模,按45 mg·kg-1的剂量一次性给大鼠左下腹腔内注射。72 h后测尾尖血血糖,血糖水平≥16.7 mmol·L-1以上且稳定3 d者为造模成功,造模成功后随机分为模型组、甲钴胺组(1.97 mg·kg-1·d-1)、白薇煎低、高剂量组(5.3,21.2 g·kg-1·d-1)ig给药,共给药4周,给药结束后,观察白薇煎对糖尿病大鼠脊神经传导速度和脊神经致痛通道蛋白的影响。结果:治疗4周后,糖尿病大鼠体质量、尾尖热敏感度和神经传导速度明显改善,脊神经致痛通道蛋白表达亦明显改变;与正常组(21.12±1.07)m·s-1比较,模型组在4周时的神经传导速度(14.23±1.13)m·s-1均有明显减慢(P<0.05),致痛通道蛋白表达高于正常组;与模型组比较,甲钴胺组在4周可明显改善神经传导速度(17.55±1.43)m·s-1(P<0.05),致痛通道蛋白表达弱于模型组,白薇煎低、高剂量组均能明显改善神经传导速度(15.49±1.36),(18.9±1.25)m·s-1(P<0.05),低剂量组的致痛通道蛋白表达略低于模型组,高剂量组的致痛通道蛋白表达弱于模型组。结论:白薇煎能改善糖尿病大鼠脊神经传导速度,降低脊神经致痛通道蛋白的表达,对糖尿病周围神经病变具有较好的防治作用。这可能是其对糖尿病周围神经病变防治作用的机制之一。 OBJECTIVE: To observe the effect of Chinese herb Bai Wei Jian on the spinal nerve conduction velocity and the protein of pain induced by spinal nerve in diabetic rats. Methods: Ten rats were randomly selected as the normal group and the rest as the model group. The rats were injected intraperitoneally with streptozotocin (STZ) at a dose of 45 mg · kg-1. After 72 h, the blood glucose was measured and the blood glucose level ≥16.7 mmol·L-1 and stable for 3 days were successfully established. After successful modeling, the rats were randomly divided into model group, mecobalamin group (1.97 mg · kg -1 · D-1), Bai Wei Jian low, high-dose group (5.3,21.2 g · kg-1 · d-1) ig administration for a total of 4 weeks after the administration, Effects of Rat Spinal Nerve Conduction Velocity and Spinal Nerve Pain Pathway Protein. Results: Compared with the normal group (21.12 ± 1.07) m s-1, the body weight, thermal sensitivity at tail tip and nerve conduction velocity of diabetic rats were significantly improved after 4 weeks of treatment, At 4 weeks, the nerve conduction velocity (14.23 ± 1.13) m · s-1 in the model group was significantly slowed down (P <0.05), and the expression of induced pain channel was higher than that in the normal group. Compared with the model group, After 4 weeks, the nerve conduction velocity (17.55 ± 1.43) m · s-1 (P <0.05) was significantly improved, and the expression of pain channel was weaker than that of model group. The effect of Bai Wei Jian low and high dose group could obviously improve the nerve conduction velocity (15.49 ± 1.36) and (18.9 ± 1.25) m · s-1, respectively (P <0.05). The protein expression of the induced channel in the low-dose group was slightly lower than that in the model group, and the expression of the induced-channel protein in the high-dose group was lower than that of the model group . Conclusion: Baiwei decoction can improve the conduction velocity of spinal nerves in diabetic rats and reduce the protein expression of the induced pain in the spinal nerves, which has a good preventive and therapeutic effect on diabetic peripheral neuropathy. This may be one of its mechanisms of prevention and treatment of diabetic peripheral neuropathy.
