以人参叶片为试材,MS为基本培养基,采用均匀设计和DPS软件分析的方法,研究了2,4-D、KT、NAA、6-BA等4种不同激素组合对人参愈伤组织诱导的影响,以期筛选出诱导人参叶片愈伤组织的最佳激素组合。结果表明:人参叶片愈伤组织诱导的最佳培养基为MS+2,4-D 2mg·L-1+NAA 0.66mg·L~(-1)+KT 1.0mg·L~(-1)。验证试验表明,在添加最佳激素组合的MS培养基中接种新的叶片外植体,30d后愈伤组织诱导率为93%,单个叶片外植体上愈伤组织小块平均数达1.82.
Using ginseng leaves as test material and MS as basic medium, the effects of 4 different hormone combinations of 2,4-D, KT, NAA and 6-BA on the induction of ginseng callus were studied by using uniform design and DPS software analysis. In order to screen out the best hormone combination that induces the callus of ginseng leaves. The results showed that the optimum culture medium for the induction of callus was: MS + 2,4-D 2mg · L-1 + NAA 0.66mg · L -1 + KT 1.0mg · L -1. The validation test showed that inoculation of new leaf explants in the MS medium supplemented with the best hormones showed that the callus induction rate was 93% after 30 days, and the average number of callus on a single leaf explant reached 1.82.
