目的检测EMA-RTQ-PCR快速鉴别沙门菌死活细胞方法的应用效果,评价该方法的实际应用价值。方法利用EMA-RTQ-PCR以及国标GB4789.4-2010对人工模拟样品及实际样品分别进行检测。结果对于人工模拟死菌样品,EMA-RTQ-PCR与国标法检测结果一致,均为阴性;对于人工模拟死活菌混合样品,EMA-RTQ-PCR直接检测结果与国标法存在差异,EMA-RTQ-PCR为阴性,国标法为阳性;人工模拟死活菌混合样品增菌后用EMA-RTQ-PCR进行检测,结果与国标法一致,均为阳性;增菌后的实际样品用EMA-RTQ-PCR进行检测的结果与国标检测结果一致,均为阴性。结论通过对样品进行前增菌,可以保证检验结果的准确可靠。该方法能够快速、灵敏、准确的检测沙门菌活细胞,具有很好的应用价值。
Objective To detect the effect of EMA-RTQ-PCR in rapid identification of dead and living Salmonella cells and to evaluate the practical value of this method. Methods EMA-RTQ-PCR and GB4789.4-2010 were used to test artificial samples and actual samples respectively. Results The result of EMA-RTQ-PCR was consistent with that of the national standard method, and the results of EMA-RTQ-PCR showed that the results of EMA-RTQ- PCR was negative, the national standard method was positive; artificial simulated live mixture of bacteria enrichment after the test by EMA-RTQ-PCR, the results are consistent with the national standard, were positive; the actual sample after enrichment by EMA-RTQ-PCR Test results and GB test results are consistent, are negative. Conclusion By pre-enrichment of the sample, you can ensure that the test results are accurate and reliable. The method can rapidly, sensitively and accurately detect live Salmonella cells and has good application value.
