为利用固定剂防止孵育时的酶扩散,有必要防止固定剂对酶活性的抑制作用。酶的底物是一种抗变剂,可增加酶蛋白的稳定性,以对抗固定剂抑制酶活性的作用。本实验选用甲醛作固定剂,固定大鼠心、肝、肾及颌下腺的新鲜组织,作恒冷切片。在固定液中分别加入G-6-Pase的底物葡萄糖-6-磷酸二钠和SDH的底物琥珀酸二钠,观察其对抗甲醛抑制G-6-Pase和SDH的作用。
It is necessary to prevent the immobilizing agent from inhibiting the enzyme activity in order to prevent the spread of the enzyme during incubation by using the fixative. The enzyme substrate is an anti-change agent that increases the stability of the enzyme protein to counter the effect of the fixative on enzyme activity. In this experiment, formaldehyde was used as fixative to fix the fresh tissues of rat heart, liver, kidney and submandibular gland for constant cold section. The substrates of G-6-Pase, glucose-6-phosphate and disodium succinate, which are the substrates of SDH, were respectively added into the fixative solution to observe their inhibitory effects on the inhibition of G-6-Pase and SDH by formaldehyde.
