目的:新生兔骨髓间充质干细胞(BMSCs)的培养方法,从各方面鉴定体外培养BMSCs的生物学特性。方法:采用贴壁筛选法纯化MSCs,镜下连续细胞的形态学观察。从形态学特征、表面标志物及其具有的向骨、脂肪分化的功能等多方面经行综合鉴定。结果:原代BMSCs呈集落状生长,细胞呈梭形、呈放射状排列。传代后大都均匀分布生长,细胞生长迅速;可向成骨细胞、脂肪细胞分化。结论:贴壁筛选法可成功分离和培养新生兔的BMSCs,BMSCs能向成骨细胞、脂肪细胞分化,可以作为组织工程中种子细胞的来源。
OBJECTIVE: To study the biological characteristics of BMSCs cultured in vitro from different aspects of culture method of newborn rabbit bone marrow mesenchymal stem cells (BMSCs). Methods: MSCs were purified by adherence screening. Morphological changes of continuous cells were observed. From the morphological characteristics, surface markers and the bone to the bone, fat differentiation and other aspects of the comprehensive identification by the line. Results: The primary BMSCs showed colony-like growth and the cells were spindle-shaped and radially arranged. After the passage of most of the uniform distribution of growth, rapid cell growth; to osteoblasts, adipocytes differentiation. Conclusion: The adherence screening method can successfully isolate and culture BMSCs of newborn rabbits. BMSCs can differentiate into osteoblasts and adipocytes, which can be used as the source of seed cells in tissue engineering.
