目的:研究复方麦芽丸(Compound Malt Pill,CMP)对多囊卵巢综合征(polycystic ovarian syndrome,PCOS)大鼠白细胞介素(IL-6)、肿瘤坏死因子(TNF-α)表达的影响,探讨复方麦芽丸治疗PCOS的作用机制。方法:运用来曲唑建立多囊卵巢综合征大鼠模型,48只6周龄雌性SD大鼠随机分为6组(n=8):正常对照组、模型对照组、达英-35 1.44g/kg组、复方麦芽丸5.76g/kg组、复方麦芽丸11.52g/kg组、复方麦芽丸23.04g/kg组。第1~21天,正常组给予1%羧甲基纤维素溶液(CMC)灌胃,其余组给予0.125mg/ml来曲唑-羧甲基纤维素溶液诱导PCOS,第22~42天,正常组和模型组给予生理盐水,其余组以相应药物灌胃,观察大鼠动情周期变化,运用免疫组化、Western blot和RT-q PCR检测各组大鼠卵巢组织中IL-6、TNF-α的表达。结果:PCOS大鼠动情周期失去规律性变化,经治疗后有所恢复;模型对照组IL-6、TNF-α的表达水平较正常组明显增高,各治疗组IL-6、TNF-α表达水平较模型组降低,尤以复方麦芽丸高剂量组(23.04g/kg)最为显著。结论:复方麦芽丸对PCOS大鼠有一定治疗作用,其机制可能是通过调节卵巢组织IL-6、TNF-α的表达水平。
Objective: To investigate the effect of Compound Malt Pill (CMP) on the expression of interleukin (IL) -6 and tumor necrosis factor (TNF-α) in polycystic ovarian syndrome (PCOS) rats The Mechanism of Compound Malting Pills in Treating PCOS. Methods: Polychondral ovarian syndrome rat model was established by letrozole. 48 6-week-old female Sprague-Dawley rats were randomly divided into 6 groups (n = 8): normal control group, model control group, / kg group, 5.76g / kg compound malt group, 11.52g / kg compound malt group and 23.04g / kg compound malt group. On day 1 to 21, 1% carboxymethylcellulose solution (CMC) was given to the normal group and the rest to 0.125 mg / ml letrozole-carboxymethylcellulose solution for PCOS induction. Results: The estrous cycle of PCOS rats lost regularity and recovered after treatment. The expression of IL-6 and TNF-α in model control group was significantly higher than that in normal control group. The expression of IL-6 and TNF-α Lower than the model group, especially the high-dose compound malt pills group (23.04g / kg) the most significant. Conclusion: Compound malt-pellet has a certain therapeutic effect on PCOS rats, which may be through the regulation of the expression of IL-6 and TNF-α in ovarian tissue.
