目的:检测正常宫颈组织和CIN及宫颈癌病灶中的CX43和Vimentin表达水平,分析二者与临床病理指标之间的关系。方法:收集2014年1月-2015年6月年高青县人民医院妇产科手术切除的33例宫颈癌、32例CIN及16例正常宫颈组织标本作为对照组,试验对象术前均未进行化疗与放疗,且均行HPV检查。采用免疫荧光与Western Blot方法检测CX43和Vimentin在3种组织中的表达水平。结果:(1)Cx43在对照组、CIN(I、II-III及宫颈癌中的阳性表达率分别为68.8%、66.7%、65%、27.3%,宫颈癌组cx43表达明显低于对照组(p=0.006);cx43在宫颈癌静脉侵犯组的表达率显著低于未侵犯组:13%VS60%,(p=0.018);在淋巴结侵犯组的表达率显著低于未侵犯组:9.1%和63.6%,(p=0.004)。(2)Vimentin在对照组、CIN(I、II-III及宫颈癌中的阳性表达率分别为12.5%、25%、50%、69.7%,进行X2检验显示阳性表达率有显著性差异(p=0.024);Vimentin在宫颈癌侵犯静脉组的表达率显著高于未侵犯静脉组:90.9%VS27.3%,(p=0.001);在宫颈癌淋巴结侵犯组的表达率显著高于淋巴结未侵犯组:86.4%和36.4%,(p=0.011)。(3)spearman相关分析显示cx43与Vimentin的表达呈负相关(r=-0.485,p=0.004)。结论:CX43表达降低和Vimentin表达增多与宫颈癌恶性程度相关,Vimentin与CX43的表达呈负相关联系。
Objective: To detect the expression levels of CX43 and Vimentin in normal cervical tissue, CIN and cervical lesions, and to analyze the relationship between them and the clinicopathological parameters. Methods: Thirty-three cases of cervical cancer, 32 cases of CIN and 16 cases of normal cervical tissue surgically removed from the department of Obstetrics and Gynecology of People’s Hospital of Gaoqing County from January 2014 to June 2015 were selected as the control group. Chemotherapy and radiotherapy, and HPV testing. Immunofluorescence and Western Blot were used to detect the expression of CX43 and Vimentin in three tissues. Results: (1) The positive expression rates of Cx43 in CIN (I, II-III and cervical cancer) were 68.8%, 66.7%, 65% and 27.3% respectively. The expression of cx43 in cervical cancer group was significantly lower than that in control group p = 0.006). The expression of cx43 in cervical cancer was significantly lower than that in noninvasive group (13% vs 60%, p = 0.018). The expression of cx43 in lymph node invasion group was significantly lower than that in noninvasive group 63.6%, (p = 0.004). (2) The positive rates of Vimentin in control group, CIN (I, II-III and cervical cancer were 12.5%, 25%, 50%, 69.7% (P = 0.024). The expression rate of Vimentin in cervical cancer invading vein group was significantly higher than that in noninvasive vein group: 90.9% VS27.3%, (p = 0.001); in cervical lymph node infiltration group (P = 0.011). (3) The spearman correlation analysis showed that the expression of cx43 was negatively correlated with the expression of Vimentin (r = -0.485, p = 0.004) .Conclusions : The decrease of CX43 expression and the increase of Vimentin expression are associated with the malignant degree of cervical cancer. The expression of Vimentin is negatively correlated with the expression of CX43.
