这是一种以刃天青(resazurin)被细菌脱氢酶还原为基础的毒性检验的生物化学方法。与常用的培养皿计数法相比,该法敏感10倍,试验仅用2小时,重复性良好,变异系数平均为10%。通过对氯化程度不同的几种氯酚及对氯酚三种异构体的试验,看出该法可适用于结构/毒性方面的研究。【试剂】刃天青溶液:BDH生产的刃天青一片(5mg/片)溶于50 ml蒸馏水。贮于棕色瓶内,在4℃条件下可稳定保存一周左右。0.05 M邻苯二酸盐-盐酸缓冲液:1.02 g
This is a biochemical method of toxicity testing based on resazurin bacterial dehydrogenase reduction. Compared with the common petri dish counting method, the method is sensitive 10 times, the test is only 2 hours, the repeatability is good, and the coefficient of variation is 10% on the average. Through the experiments on three isomers of chlorophenols and p-chlorophenols with different degrees of chlorination, we can see that this method can be applied to the study of structure / toxicity. [Reagents] Resazurin solution: BDH production of resazurin (5mg / tablet) dissolved in 50 ml of distilled water. Stored in a brown bottle, at 4 ℃ under the conditions of stable storage for about a week. 0.05 M Phthalate-hydrochloric acid buffer: 1.02 g
