目的对安徽省首例人感染H5N6禽流感病例相关标本进行实验室检测,为今后禽流感疫情实验室检测和防控提供有效参照。方法采用荧光实时定量PCR(real-time PCR)反应检测疑似病例和外环境标本以及密切接触者的咽拭子标本,对NA基因片段采用一步法普通RT-PCR技术进行扩增。将阳性外环境标本和疑似病例标本送安徽省疾病预防控制中心流感参比中心实验室复核。结果疑似病例咽拭子标本经实验室检测为H5N6亚型流感病毒。在13份农贸市场标本中,有2份H5N6和H9N2亚型混合标本和1份H5N6标本,5份H9N2亚型标本,1份H7亚型标本,1份H7和H9N2混合标本。其他标本均阴性。结论经宣城市疾病预防控制中心、安徽省疾病预防控制中心和中国疾病预防控制中心病毒所国家流感中心一致鉴定,该患者为安徽省首例人感染H5N6高致病性禽流感病例。
Objective To carry out laboratory tests on the first case of H5N6 bird flu in Anhui Province and provide a valid reference for future laboratory testing and prevention and control of bird flu. Methods Fluorescent real-time PCR (real-time PCR) was used to detect throat swab specimens from suspected and extrinsic environmental samples and close contacts. The NA gene fragments were amplified by one-step RT-PCR. Positive specimens of the outer environment and suspected cases were sent to the Center for Disease Control and Prevention in Anhui Province Center for Reference Control influenza review. Results Suspected cases of throat swab specimens tested by the laboratory as H5N6 subtype influenza virus. Among the 13 farmer’s market samples, there were 2 mixed samples of H5N6 and H9N2 subtypes, 1 H5N6 sample, 5 H9N2 subtype samples, 1 H7 subtype sample and 1 mixed sample of H7 and H9N2. Other specimens were negative. Conclusion The cases were identified by the National Influenza Center of Xuancheng Center for Disease Control and Prevention, Anhui Provincial Center for Disease Control and Prevention and China National Center for Disease Control and Prevention for the first human case of H5N6 HPAI in Anhui Province.
