Different mannose concentration grades were designed to detect the tolerance of sugarcane varieties ROC22 and GT21 during stages of callus induction,differentiation and rooting.The results showed that the two sugarcane varieties were sensitive to the mannose,and the optimum concentrations of mannose at different stages were determined,correspondingly.Mannose positive selection system of sugarcane was developed,and the efficient transformation system of ROC22 and GT21 mediated by Agrobacterium tumefaciens was established.The pmi gene has been integrated into the sugarcane genome which was authenticated by PCR and chlorophenol red sodium(CPR)detection as indicated by 30.0 and 37.8%transformation frequency,respectively.
