Intrahepatic expression of hepatitis B x antigen (HBxAg) is associated with the development of HCC, perhaps through up-regulated expression of selected cellular genes.Accordingly, HepG2 cells were stably transduced with recombinant retrovirus making HBxAg or the bacterial chloramphenicol acetyltransferase (CAT) product.RNA isolated from HepG2X and HepG2CAT cells was subjected to PCR select cDNA subtraction.Differentially expressed genes were validated in clinical samples by in situ hybridization, northern and western blotting, and by immunohistochemistry.Following validation, selected clones were functionally characterized in liver cell culture.
