Cellular uptake and endosomal escape are major barriers to gene delivery.The arginine-rich TAT peptides are able to efficiently translocate exogenous DNA across the plasma membrane via endocytosis.Unfortunately,molecules taken up by endocytosis undergo endosomal entrapment and possible metabolic degradation.To overcome the critical barriers to gene delivery,we designed and tested a series of multifunctional peptides.These peptides contained a cationic amphiphilic α-helical antimicrobial peptide(AMP)fragment,and a stearylated cell-penetrating peptide(CPP)TAT with an insertion of cysteine for more compact condensation of DNA.The peptides(G(LLKK)3G-TAT,C(LLKK)3C-TAT,C18-G(LLKK)3G-TAT and C18-C(LLKK)3C-TAT),which contained different functional fragments,were evaluated for their ability to mediate gene delivery in 293T cells and NIH-3T3 cells.
