应用接触电极在位法、悬浮式微电极在位法和离体心肌组织灌流法观察了参附注射液对家兔心肌细胞的电生理效应。结果表明,三种方法下,参附液均能显著增加动作电位Vmax(接触电极法增加41.84%、悬浮法增加30.35%、离体法增加30.33%)。而对动作电位振幅(APA)和动作电位时程(APD)影响很小。同时发现参附液能部份对抗河豚毒素(TTX)的快Na~+通道阻滞作用,对细胞具有保护作用。通过对参附液效应的比较研究,作者对三种方法在电生理研究中的应用价值进行了初步评价。
The electrophysiological effects of Shenfu injection on myocardial cells in rabbits were observed by contact electrodeposition method, suspended microelectrode in situ method and isolated myocardial tissue perfusion method. The results showed that, under the three methods, Shenfu solution could significantly increase the action potential Vmax (41.84% increase in contact electrode method, 30.35% increase in suspension method, and 30.33% increase in in vitro method). However, the effect of action potential amplitude (APA) and action potential duration (APD) is small. At the same time, it was found that Shenfu liquid can partially block the fast Na~+ channel blockade effect of tetrodotoxin (TTX), and has a protective effect on cells. Through the comparative study of the effects of the paraffin-attached fluid, the author made a preliminary evaluation of the application value of the three methods in the electrophysiological study.
