c-Fms是原癌基因fms的产物,是酪氨酸激酶受体家族成员之一,与单核细胞/巨噬细胞的增殖、分化及其活性相关。c-Fms的胞外区含有5个Ig样结构域,胞内区具有酪氨酸激酶活性,是具有单一高亲和力受体分子。M-CSF与c-Fms Ig样结构域的D1、D2和D3结合,引起D4构象变化、受体二聚化而导致胞内区酪氨酸激酶的活化。它的功能涉及STATs、JAK、Ras、Raf-1、MEK、MAPK以及PI-3K、PKC等。其底物是一些含有SH2结构域的蛋白质分子,逐个活化这些蛋白质分子,进而调控c-myc、c-fos和c-jun等相关基因的表达,实现受体信号在细胞内传递。
c-Fms, a product of the oncogene fms, is a member of the tyrosine kinase receptor family and is associated with the proliferation, differentiation and activity of monocytes / macrophages. The extracellular domain of c-Fms contains 5 Ig-like domains, the intracellular region has tyrosine kinase activity and is a single high-affinity receptor molecule. M-CSF binds to D1, D2 and D3 of the c-Fms Ig-like domain, causing a conformational change in D4 that leads to the activation of intracellular tyrosine kinases by receptor dimerization. Its function involves STATs, JAK, Ras, Raf-1, MEK, MAPK and PI-3K, PKC and so on. Its substrate is a number of SH2 domain containing protein molecules, one by one activation of these protein molecules, and then regulate the expression of c-myc, c-fos and c-jun and other related genes, to achieve the receptor signal transmission in the cell.
