应用自动化图像分析仪对低剂量α-双炔失碳酯(ANO)诱导HL-60细胞分化的作用进行了形态测量研究。4μg/mlANO连续作用HL-60细胞5天。共测8个形态指标。结果表明,细胞面积:核面积,核周长,核最大直径,核最小直径,核灰度/核面积随药物作用时间增加而增大,而核浆比却随时间增加而减小。所有数据表明,在低剂量ANO(4μg/ml)作用下,HL-60细胞在1~4天内作为细胞生命基本的DNA变化显著,但细胞形态基本稳定。第5天开始呈现显著性地向成熟细胞转化的倾向。
The morphometric study of the effects of low-dose α- anordyne (ANO)-induced differentiation of HL-60 cells was performed using an automated image analyzer. HL-60 cells were continuously treated with 4 μg/ml ANO for 5 days. A total of 8 morphological indicators were measured. The results showed that the cell area: nuclear area, nuclear perimeter, maximum nuclear diameter, nuclear minimum diameter, nuclear grayscale/nuclear area increased with the increase of drug action time, but the nuclear-to-plasma ratio decreased with time. All data showed that under low-dose ANO (4 μg/ml), HL-60 cells showed significant DNA changes in cell life within 1 to 4 days, but cell morphology was basically stable. Day 5 began to show a significant tendency to transform into mature cells.
