目的:研究补阳还五汤对局灶性脑缺血大鼠血管生成和神经功能恢复的影响。方法:采用线栓法诱导大鼠大脑中动脉阻塞,缺血90min后再灌,缺血后24h开始灌胃补阳还五汤,1天1次,连续14天,在缺血后第1、3、7、14天分别采用肢体放置实验、错步实验和粘胶揭除实验评价感觉运动功能;采用免疫组化检测缺血周边区微血管密度(microvessels density,MVD)和血管内皮生长因子(vascular endothelial growth factor,VEGF)表达。结果:与模型组比较,补阳还五汤不仅能显著提高肢体放置实验成绩,减少错步次数和缩短粘胶揭除潜伏期(P<0.05或P<0.01),而且能显著增加缺血周边区vWF阳性血管和VEGF阳性细胞数量(P<0.01)。结论:补阳还五汤能促进脑缺血后神经功能恢复,机制可能与其促进缺血周边区血管生成和VEGF表达有关。
Objective: To investigate the effects of Buyang Huanwu Decoction on the angiogenesis and neurological recovery after focal cerebral ischemia in rats. Methods: The middle cerebral artery occlusion was induced in rats by thread suture method. The rats were re-filled with blood for 90 minutes after ischemia, and beihuanghuwu decoction was infused for 24 hours after ischemia. The rats were sacrificed for 1 day, At 3, 7 and 14 days, the sensory motor function was assessed by limb placement test, misdiagnosis test and mucilage removal test, respectively. The expression of microvessels density (MVD) and vascular endothelial growth factor (vascular endothelial growth factor) endothelial growth factor (VEGF)). Results: Compared with the model group, Buyang Huanwu Decoction could not only significantly improve the experimental results of limb placement, reduce the number of steps and shorten the period of removing the adhesive (P <0.05 or P <0.01), but also significantly increase the ischemic peripheral area vWF-positive vessels and VEGF-positive cells (P <0.01). Conclusion: Buyang Huanwu decoction can promote the recovery of neural function after cerebral ischemia, the mechanism may be related to promoting angiogenesis and VEGF expression in ischemic border area.
