目的检测胃癌组织和癌旁组织中肿瘤干细胞标志物CD133的表达,探讨其与临床病理参数的相关性和在肿瘤诊断中的意义。方法收集36例胃癌患者手术切除的癌组织和对应的癌旁组织,用RT-PCR检测CD133的mRNA表达,用免疫组织化学技术检测CD133蛋白质的表达,分析CD133 mRNA在胃癌组织中的表达与临床病理参数的相关性。结果 CD133 mRNA表达阳性率为胃癌组织66.7%(24/36),癌旁组织为16.7%(6/36),差异有统计学意义(P<0.01);胃癌组织中CD133表达阳性率与肿瘤分化状态相关,低、未分化组高于中、高分化组(P<0.01),但与年龄、性别、肿瘤大小、浸润深度、TNM分期和有无淋巴结转移无关。结论肿瘤干细胞标志物CD133的表达与胃癌的发生及分化状态相关,可作为胃癌诊断的一个新的分子标志物。
Objective To detect the expression of tumor stem cell marker CD133 in gastric cancer tissues and adjacent non-cancerous tissues, and to explore its correlation with clinical pathological parameters and its significance in tumor diagnosis. Methods Thirty-six gastric cancer tissues and corresponding para-cancerous tissues were collected. The mRNA expression of CD133 was detected by RT-PCR. The expression of CD133 protein was detected by immunohistochemistry. The expression of CD133 mRNA in gastric cancer tissues was analyzed. Correlation of pathological parameters. Results The positive rate of CD133 mRNA expression in gastric cancer was 66.7% (24/36) and that in paracancerous tissues was 16.7% (6/36), the difference was statistically significant (P <0.01). The positive rate of CD133 in gastric cancer was correlated with tumor differentiation However, it was not related to age, sex, tumor size, depth of invasion, TNM staging and lymph node metastasis (P <0.01). Conclusion The expression of tumor stem cell marker CD133 correlates with the occurrence and differentiation of gastric cancer and may be used as a new molecular marker for the diagnosis of gastric cancer.
