分离、培养佐剂性关节炎(AA)大鼠膝关节滑膜细胞,倒置相差显微镜观察细胞形态,检测细胞增殖能力及培养上清中白细胞介素1(IL-1)、肿瘤坏死因子(TNF-α)的生物活性和前列腺素E2(PGE2)的含量。结果表明AA大鼠滑膜细胞存在三种不同的形态,即巨噬细胞样细胞(MCs)、树突状细胞(DCs)、成纤维细胞样细胞(FCs);滑膜细胞增殖能力增强;其分泌IL-1、TNF-α和PGE2的能力增强。本实验为研究类风湿性关节炎的病理及筛选抗风湿药提供了理想的实验材料。
The synoviocytes of knee joints of adjuvant arthritis (AA) rats were isolated and cultured. The morphological changes of cells were observed under inverted phase contrast microscope. The proliferation of cells and the expression of interleukin 1 (IL-1), tumor necrosis factor (TNF) -α) and prostaglandin E2 (PGE2) content. The results showed that there are three different morphological changes of AA rat synoviocytes: macrophage-like cells (MCs), dendritic cells (DCs) and fibroblastoid cells (FCs) The ability to secrete IL-1, TNF-α and PGE2 is enhanced. This experiment for the study of rheumatoid arthritis pathology and screening anti-rheumatic drugs provides an ideal experimental material.
