在许多作物中,利用生长点组织培养,已可达到无病毒化并进入大量繁殖的应用阶段。1978年在日本首次发现莲藕病毒病。考虑到莲藕是营养繁殖的作物,因而,将来有必要进行无病毒苗生产或试管优良种苗的大量繁殖。本研究旨在建立一种以莲藕无病毒化植株及其大量增殖方法为基础的生长点组织培养方法。 供试材料为白花莲(Nelumbo nucifera)。切取生育期植株地下茎顶部15—20毫米,用70%酒精和稀释20倍的次氯酸钠(有效氯0.25%)溶液各进行30秒和30分钟的杀菌处理,然后用无菌水洗净。接着,在实体显微镜下取下带有1个叶原基的顶芽或侧芽
