目的观察妊娠期小鼠暴露尼古丁对后代大脑皮层和海马形态结构发育及对神经细胞黏附分子(NCAMs)表达的影响。方法建立妊娠期暴露尼古丁的小鼠动物模型,HE染色观察后代大脑皮层和海马形态结构的改变,免疫组织化学法、蛋白质免疫印迹法观察生后小鼠NCAMs表达的变化。结果妊娠期暴露尼古丁小鼠的后代其大脑皮层和海马变薄,海马各区细胞层也变薄。与对照组相比,妊娠期暴露尼古丁小鼠的后代大脑皮层和海马中,PSA-NCAM表达升高,而NCAM-180、140和120的表达减弱。结论妊娠期小鼠暴露尼古丁能阻碍后代大脑皮层和海马形态结构的正常发生,还能改变NCAMs在生后小鼠大脑皮层和海马中的表达。
Objective To observe the effect of nicotine exposure on the morphological and structural changes of cortex and hippocampus and the expression of neural cell adhesion molecules (NCAMs) in the offspring of pregnant mice. Methods The animal model of nicotine exposed during pregnancy was established. The morphological changes of cerebral cortex and hippocampus were observed by HE staining. The expression of NCAMs in postnatal mice was observed by immunohistochemistry and Western blotting. Results Offspring of gestational nicotine-exposed mice became thinner in the cerebral cortex and hippocampus, and the cell layers in the hippocampus also became thinner. Compared with the control group, the expression of PSA-NCAM was increased in the cortex and hippocampus of offspring of gestational nicotine-exposed mice, while the expression of NCAM-180, 140 and 120 was weakened. Conclusion Exposure of nicotine to mice during pregnancy inhibits the normal morphology of the cerebral cortex and hippocampus in the offspring and changes the expression of NCAMs in postnatal mouse cerebral cortex and hippocampus.
