同型半胱氨酸/半胱氨酸是体内的含硫氨基酸,近年来提出它们可以诱导血管平滑肌细胞的增殖,是心血管病的一个新的独立的危险因子.但其机制尚不了解,本研究目的在于克隆同型半胱氨酸/半胱氨酸诱导的新基因.应用差异显示即RNAmapping的方法,从半胱氨酸诱导的大鼠血管平滑肌细胞中,寻找有差异的条带,克隆新的cDNA.结果分离出7种上调和4种下调cDNA,并且发现其中有4个是至今尚未发现的新的cDNA.应用Northernblot分析,这4个新的mRNA其长度分别为5.0、2.0、1.8和2.2kb.它们可能在血管平滑肌细胞增殖和心血管病的发病中具有重要意义.
Homocysteine / cysteine, the sulfur-containing amino acids in the body, has been proposed in recent years to induce proliferation of vascular smooth muscle cells and is a new, independent risk factor for cardiovascular disease. However, its mechanism is still unknown. The purpose of this study was to clone a novel homocysteine / cysteine-induced gene. Using RNAmapping, a differential display method, we searched for different bands from cysteine-induced rat vascular smooth muscle cells and cloned the new cDNA. As a result, seven kinds of up-regulation and four kinds of down-regulation cDNAs were isolated, and four of them were found to be new cDNAs that have not been discovered yet. Northernblot analysis showed that the four new mRNAs had lengths of 5.0, 2.0, 1.8 and 2.2 kb, respectively. They may play an important role in the proliferation of vascular smooth muscle cells and the pathogenesis of cardiovascular diseases.
