本文报道用二种显色反应能选择性地鉴定和测定5-氟尿嘧啶和尿嘧啶氮芥二种尿嘧啶抗癌药物制剂。 5-氟尿嘧啶在硼砂介质中用溴水处理,然后在酸性介质中与2,4-二硝基苯肼生成一种橙红色沉淀,用氢氧化钾溶液处理后,产生明显紫色。含有烷化基的尿嘧啶氮芥,与α-皮考林水溶液共热,所形成的季铵盐在碱性介质中与1,2-萘醌-4-磺酸钠缩合,产生粉红色。这些颜色反应能用于测定纯品和药物制剂中的这些抗癌药物含量。5-氟尿嘧啶和尿嘧啶氮芥含量分别在2.5~5μg/ml和5~25μg/ml的浓度范围内符合Beer’s定
This article reports two kinds of chromogenic reaction can be selectively identified and determination of 5-fluorouracil and uracil mustard two uracil anticancer drug formulations. 5-Fluorouracil was treated with brominated water in borax medium and then with orange 2,4-dinitrophenylhydrazine in an acidic medium to produce an orange-red precipitate which, after treatment with potassium hydroxide solution, produced a clear purple color. The uracil mustard, which contains an alkylating group, is co-heated with the a-picoline solution and the resulting quaternary ammonium salt is condensed with 1,2-naphthoquinone-4-sulphonate in an alkaline medium to produce a pink color. These color reactions can be used to determine the content of these anti-cancer drugs in both pure and pharmaceutical formulations. The concentrations of 5-Fluorouracil and uracil mustard were within the range of 2.5 ~ 5μg / ml and 5 ~ 25μg / ml respectively
