目的考察大鼠灌服芎汤有效部位后,有效成分阿魏酸在大鼠体内排泄动力学规律。方法芎汤醇提液经分离制备的有效部位给SD大鼠灌胃给药后,按不同时间段收集大鼠的尿液和粪样,对其中阿魏酸进行HPLC测定,建立其尿液和粪样排泄代谢曲线,考察排泄特征。结果尿液和粪样中阿魏酸的半衰期分别为(3.37±1.2)h、(23.09±5.7)h;排泄速率常数分别为(0.21±0.002)h-1、(0.03±0.0005)h-1。结论阿魏酸在尿液和粪便中的累积排泄率为11.77%,说明小部分阿魏酸以原型药通过粪便和尿液排泄,大部分阿魏酸在大鼠体内转化为其他形式。该研究为阐明阿魏酸体内过程提供一定依据。
Objective To investigate the regularity of excretion of ferulic acid in rats after the rats were ingested with the effective part of Chuanxiong Decoction. Methods Xiong-tao Tang alcohol extract prepared by the effective part of the SD rats after gavage administration, according to different time periods to collect urine and fecal samples of rats, ferulic acid HPLC determination of its urine Excretion and metabolism of fecal samples, excretion characteristics. Results The half-lives of ferulic acid in urine and fecal samples were (3.37 ± 1.2) h and (23.09 ± 5.7) h respectively, and the excretion rate constants were (0.21 ± 0.002) h-1 and (0.03 ± 0.0005) h- . Conclusions The cumulative excretion rate of ferulic acid in urine and feces was 11.77%, indicating that a small amount of ferulic acid was excreted in excrement and urine by the prototype drug. Most ferulic acid was converted to other forms in rats. This study provides some evidence to elucidate the process of ferulic acid in vivo.
