目的:研究藻蓝蛋白对2型糖尿病大鼠心脏形态结构和心肌细胞凋亡的影响。方法:健康雄性Wistar大鼠32只,高脂高糖饲料喂养联合小剂量链脲佐菌素(STZ)腹腔注射方法建立2型糖尿病大鼠模型,藻蓝蛋白灌胃治疗,组织病理学技术观察心脏的形态结构,用原位末端标记法(Tunel法)检测心肌细胞凋亡。结果:正常组大鼠心肌纤维走行和结构正常,心脏中均可见少量散在的棕黄色染色的凋亡阳性心肌细胞;模型组大鼠心肌纤维肥大、颗粒变性、脂肪变性,凋亡阳性细胞较正常组明显增多,凋亡指数有显著差异(P<0.05);藻蓝蛋白组和二甲双胍组心肌细胞形态结构较模型组显著改善,正常细胞数目增多、排列较规则,肿胀和变性细胞较模型组显著减少,凋亡阳性细胞较模型组显著减少,凋亡指数有显著差异(P<0.05)。藻蓝蛋白组和二甲双胍组凋亡指数无显著性差异(P>0.05)。结论:藻蓝蛋白可抑制糖尿病大鼠心肌细胞凋亡,发挥一定的保护作用。
Objective: To study the effects of phycocyanin on cardiac morphology and cardiomyocyte apoptosis in type 2 diabetic rats. Methods: Twenty-two healthy male Wistar rats were induced by high fat and high glucose diet combined with low dose streptozotocin (STZ) intraperitoneal injection to establish type 2 diabetic rat model. The therapeutic effect of phycocyanin was observed by histopathology The morphological structure of the heart, cardiomyocyte apoptosis was detected by Tunel method. Results: The myocardial fibers of normal rats walked and their structures were normal. A small amount of scattered brownish brown apoptotic cardiomyocytes were seen in the heart. The myocardial fibrosis, granular degeneration, steatosis and apoptotic positive cells in model group were more than normal (P <0.05). The morphology of cardiomyocytes in phycocyanin group and metformin group was significantly improved compared with model group, the number of normal cells increased, the arrangement was more regular, and the number of swelling and degeneration cells was significantly higher than that of model group Compared with model group, the number of apoptotic positive cells decreased significantly (P <0.05). There was no significant difference in apoptosis index between phycocyanin group and metformin group (P> 0.05). Conclusion: Phycocyanin can inhibit cardiomyocyte apoptosis in diabetic rats and play a protective role.
