为了探讨褪黑素对肉羊体外胚胎生产效果的影响,本实验体外成熟液(TCM199),受精液及胚胎培养液(SOF)添加不同浓度(0、10~(-8)、10~(-9)、10~(-10))褪黑素,研究在体外胚胎生产全程添加褪黑素对体外受精效果的影响。本实验分别从屠宰场采集肉羊卵巢获得卵母细胞,卵母细胞经体外成熟、体外受精、早期胚胎体外培养。试验结果表明:体外胚胎生产过程中添加10~(-9)褪黑素组的卵母细胞卵裂率、囊胚率及囊胚细胞数显著优于其对照组(P<0.05);说明体外胚胎生产过程中全程添加抗氧化剂,可以达到抗细胞过氧化反应的目的,克服早期胚胎的发育阻滞,提高体外胚胎的囊胚发育率及囊胚细胞数,提高囊胚质量。
In order to investigate the effect of melatonin on the embryo production in vitro, the effect of melatonin on the embryo production in vitro was studied. The effects of different concentrations (0,10 ~ (-8), 10 ~ (-9) ), 10 ~ (-10)) melatonin, to study the effect of adding melatonin on in vitro fertilization effect in vitro embryo production. In this experiment, sheep ovaries were collected from slaughterhouse to obtain oocytes, oocytes matured in vitro, in vitro fertilization, early embryo culture in vitro. The results showed that the oocyte cleavage rate, blastocyst rate and blastocyst number of 10 ~ (-9) melatonin group were significantly higher than that of control group (P <0.05) in vitro during in vitro embryo production; During the whole process of embryo production, adding anti-oxidant can achieve the purpose of anti-cell peroxidation, overcome the retardation of early embryo, increase the rate of blastocyst and the number of blastocyst in vitro, and improve the quality of blastocyst.
