目的 观察γ射线照射后小鼠脾赃淋巴细胞凋亡的变化规律与Fas(CD95)表达的关系。方法 采用PI染色、双荧光流式细胞术及免疫组织化学的ABC方法研究不同剂量γ射线照射后不同时间,小鼠脾脏淋巴细胞凋亡及CD95表达的规律。结果 小鼠脾脏淋巴细胞对不同剂量照射的凋亡敏感性有显著性差异,4Gy照射后凋亡率最高(在2~6Gy之间观察),淋巴细胞表面Fas的表达随照射后细胞凋亡率的变化而改变。结论 B细胞较T细胞具有更高的辐射凋亡敏感性;脾脏淋巴细胞在辐射后表现出明显的Fas介导凋亡。
Objective To investigate the relationship between the changes of spleen lymphocyte apoptosis and the expression of Fas (CD95) in mice after γ-ray irradiation. Methods PI staining, double fluorescence flow cytometry and immunohistochemical ABC method were used to study the rules of splenic lymphocyte apoptosis and CD95 expression at different time after γ-ray irradiation. Results The sensitivity of mouse spleen lymphocytes to different doses of irradiation was significantly different. The apoptosis rate of 4Gy after irradiation was the highest (observed between 2 and 6 Gy), and the expression of Fas on the lymphocytes was increased with the apoptosis rate Change and change. Conclusion B cells have higher radiation-induced apoptosis sensitivity than T cells. Spleen lymphocytes show significant Fas-mediated apoptosis after irradiation.
