凋亡细胞在正常肝组织、肝硬变组织及肝细胞癌中具有不同的空间分布特点.定性研究表明,在正常肝组织和肝细胞癌,调亡细胞呈散在分布,在肝硬变组织主要位于假小叶周边近纤维隔处,形成“凋亡细胞带”.为了更进一步地阐明凋亡细胞在不同病理情况下的生物学意义,本文运用分形几何理论,对凋亡细胞特定的空间分布进行分数维数(分维)定量.利用原位脱氧核糖核酸末端转移酶标记技术在10例正常肝、10例肝硬变和10例肝细胞癌组织中进行原位细胞凋亡标记.凋亡细胞计数主要按Naeim F报道(1996)的方法,即在Eagle SightⅡ型图像分析系统上,将切片上的显微图象转化为灰度数字图象,在此图象上,套上四个同心圆,其最内层圆半径为62.4μm,圆面积向外依次增加一位,至最外层圆半径为176.5μm,分别计数四个圆内的凋亡细胞数;每个标本计数四个区域,分维按公式Xc.c.r~D计算,其中X为凋亡细胞数,r为圆的半径,D为分维;用X与r的对数作图,该图的斜率即为D.D值如接近2,表明分布较均匀,D值越远离2,说明分布越不均匀.结果显示,凋亡细胞体积较小,细胞核染色质浓缩,阳性信号均位于细胞核.凋亡细胞密度在正常肝组织为193.2个/mm~2,肝细胞癌组织为106.3个/mm~2,肝硬变组织为245.4个/mm~2.凋亡细胞分布的分维在正常肝组织为1.814,肝细胞癌
