目的:观察蟾蜍毒素提取物对白血病K562细胞杀伤作用及对细胞周期的影响。方法:用无水乙醇、二甲基亚砜(DMSO)、PBS缓冲液浸溶中华大蟾蜍的耳后腺及皮下腺分泌物,采用台盼蓝拒染法检测细胞增殖抑制作用,流式细胞仪检测对细胞周期的影响。结果:3种浸液均能抑制K562细胞的增殖,其抑制强度依次为乙醇浸液>DMSO浸液>PBS浸液。5 ng/ml的乙醇浸液、50 ng/ml的DMSO浸液、500 ng/ml的PBS浸液均能使K562细胞发生G2/M期阻滞。结论:蟾蜍毒素粗提物对白血病K562细胞增殖具有明显的抑制作用,并伴有G2/M期阻滞。
Objective: To observe the killing effect of toad toxin extract on leukemia K562 cells and its effect on cell cycle. Methods: The secretions of posterior gland and subcutaneous gland of Bufo gargarizans were digested with absolute ethanol, dimethylsulfoxide (DMSO) and PBS buffer. The inhibitory effect on cell proliferation was detected by trypan blue exclusion staining. Flow cytometry Instrument detection on the cell cycle. Results: The three kinds of immersion liquid could inhibit the proliferation of K562 cells. The inhibitory strength of the three kinds of immersion liquid were ethanol extract> DMSO extract> PBS extract. The G2 / M arrest of K562 cells was induced by 5 ng / ml ethanol, 50 ng / ml DMSO and 500 ng / ml PBS. Conclusion: Toad toxin extract has a significant inhibitory effect on the proliferation of leukemia K562 cells with G2 / M phase arrest.
