目的:观察中药清化散对创伤性异位骨化模型大鼠骨形态发生蛋白-4(BMP-4)的影响。方法:80只SD大鼠,除16只作为正常对照组外,其余64只行右侧跟腱切断术,建立创伤性异位骨化大鼠模型,造模后第18天,除正常对照组外,将造模后大鼠随机分为4组,分别是模型组、局部外用清化散治疗组、口服西乐葆治疗组、局部外用清化散并口服西乐葆治疗组,分别给予相应的药物治疗。用药32、61天后每组各处死一半大鼠,取组织标本,检测跟腱组织内BMP-4的含量。结果:用药32天后,模型组的BMP-4含量明显高于正常对照组(P<0.01)和治疗组(P<0.05)。各治疗组的BMP-4含量均较对照组稍高,但无显著性差异(P>0.05),各治疗组之间也无显著性差异(P>0.05)。用药61天后,模型组的BMP-4含量较30天前增幅更大,明显高于正常对照组(P<0.01)和治疗组(P<0.05)。各治疗组的BMP-4含量较30天前降低,接近正常对照组,与对照组之间无显著性差异(P>0.05)。结论:中药清化散、西乐葆或者二者联合应用均能抑制跟腱组织内BMP-4的合成,从而抑制创伤性异位骨化的发生。
Objective: To observe the effect of Qinghua Powder on bone morphogenetic protein-4 (BMP-4) in rats with traumatic heterotopic ossification. There was no significant difference between each treatment group (P> 0.05). After 61 days, the content of BMP-4 in the model group increased more than that in the 30 days before treatment, which was significantly higher than that in the normal control group (P <0.01) and the treatment group (P <0.05). The content of BMP-4 in each treatment group was lower than that of 30 days ago, close to the normal control group, and there was no significant difference (P> 0.05) with the control group. Conclusion: The combination of Qinghua Powder, Celebrex, or both can inhibit the synthesis of BMP-4 in Achilles tendon tissue and thus inhibit the occurrence of traumatic heterotopic ossification.
