本文对小鼠肺腺癌细胞系(LA-795)进行单细胞克隆化培养,分离;经筛选获得具有裸小鼠体内移植自发肺转移力相异的4株克隆化亚系,说明母系确实存在转移潜能的异质性。体外实验比较此四株亚系的结果显示:癌细胞的转移潜能不仅与其细胞增殖、分裂及胞浆骨架分布的不均、紊乱程度有关(P<0.01),而且与其胞浆骨架聚集于胞核一侧及内源性非特异性酯酶的活性表达有关(P<0.01):同时,高转移潜能亚系比低转移潜能亚系出现较低的移动性和粘连性(P<0.01)。提示:癌细胞的胞浆骨架分布不均、紊乱、聚集于胞核一侧及内源性非特异性酯酶的高表达,可能是高转移性癌细胞的重要生物学表型;癌细胞的体内转移力不一定与其体外移动性、粘连性呈正相关。
In this study, single cell clone culture and isolation of mouse lung adenocarcinoma cell line (LA-795) were carried out. Four clonal subfunctions with different spontaneous lung metastases in vivo were obtained by screening, Heterogeneity of metastatic potential. The results of in vitro experiments comparing these four sublines showed that the metastatic potential of cancer cells was not only related to the proliferation and division of cells, but also to the distribution of cytoplasmic skeleton (P <0.01), but not with the cytoskeleton (P <0.01): At the same time, the high metastatic potential line showed lower mobility and blocking than the low metastatic potential line (P <0) .01). It is suggested that the cytoplasmic framework of cancer cells are unevenly distributed, disorganized, highly concentrated on the side of the nucleus and endogenous non-specific esterase, which may be an important biological phenotype of highly metastatic cancer cells. The transfer force is not necessarily positively related to its in vitro mobility and adhesiveness.
