目的建立大鼠牙龈炎、正畸牙移动实验模型,研究正畸牙移动对正常牙周组织与炎性牙周组织的改建中血管内皮生长因子(VEGF)的表达和分布。方法选取健康雄性Wistar大鼠55只,6~8周龄,体重(220±10)g,随机分为三组,空白对照组5只不做任何处理;正常加力组25只(对照组);炎性加力组25只(实验组),加力后1、3、7、14和21d观察大鼠牙周组织VEGF的表达。结果实验组中VEGF的阳性表达高于对照组,VEGF在对照组中的表达第十四天到达峰值,在实验组中第七天到达峰值。结论 VEGF在牙周组织改建中起到重要作用,牙龈的炎症刺激和正畸牙移动所引起牙周组织改建均引起VEGF的表达变化。适宜的正畸力对伴有炎症的牙周组织的改建没有破坏作用且有助于牙周组织的改建。
Objective To establish an experimental model of gingivitis and orthodontic tooth movement in rats and study the expression and distribution of vascular endothelial growth factor (VEGF) in normal periodontal tissue and inflammatory periodontal tissue during orthodontic tooth movement. Methods Fifty-five healthy male Wistar rats aged 6-8 weeks and weighing 220 ± 10 g were randomly divided into three groups. Five rats in the blank control group received no treatment. 25 rats in the normal group (control group) ; 25 rats in experimental group (experimental group), and the expression of VEGF in periodontal tissues of rats at 1, 3, 7, 14 and 21 days after mechanical stress. Results The positive expression of VEGF in the experimental group was higher than that in the control group. The expression of VEGF in the control group reached its peak on the fourteenth day and peaked on the seventh day in the experimental group. Conclusions VEGF plays an important role in the reconstruction of periodontal tissue. The expression of VEGF is induced by the inflammation of the gingival and the periodontal tissue remodeling caused by orthodontic tooth movement. Proper orthodontic force has no disruptive effect on the reconstruction of periodontal tissue with inflammation and contributes to the reconstruction of periodontal tissue.
