目的采用乙酰丙酮分光光度法检测亚单位流感疫苗中的甲醛含量。方法采用乙酰丙酮分光光度法检测亚单位流感疫苗中的甲醛含量,对测定波长、乙酰丙酮、乙酸、乙酸铵浓度、保温时间、降温时间进行优化,并对方法进行验证。结果在500μl样品和4.5 ml乙酰丙酮溶液反应体系中,在0.125%乙酰丙酮、1.5%乙酸、25%乙酸铵条件下,40℃保温30 min,室温静置30 min后,在波长414 nm处测定甲醛含量,可获得最佳检测效果;在1~50μg/ml的浓度范围内标准曲线线性关系良好,回收率为101.9%,最低定量限为1μg/ml。结论应用乙酰丙酮分光光度法检测亚单位流感疫苗中的甲醛含量简便、快速、准确,灵敏度高,适用于实验室对甲醛含量的常规检测。
Objective To detect the content of formaldehyde in subunit influenza vaccine by acetylacetone spectrophotometry. Methods The formaldehyde content of subunit influenza vaccine was detected by acetylacetone spectrophotometry. The determination of wavelength, acetylacetone, acetic acid, ammonium acetate concentration, holding time and cooling time were optimized, and the method was validated. Results The reaction was incubated at 40 ℃ for 30 min under the conditions of 0.125% acetylacetone, 1.5% acetic acid and 25% ammonium acetate in 500 μl sample and 4.5 ml acetylacetone solution. After standing at room temperature for 30 min, the absorbance was measured at 414 nm Formaldehyde content, the best detection effect can be obtained. The standard curve has a good linearity within the range of 1 ~ 50μg / ml with the recovery of 101.9% and the lowest limit of quantitation of 1μg / ml. Conclusion The method of acetylacetone spectrophotometry for the determination of formaldehyde in subunit influenza vaccine is simple, rapid, accurate and sensitive. It is suitable for the routine testing of formaldehyde in laboratory.
