目的探讨内蒙地区汉族冠心病患者的ACE基因I/N的多态性,并同汉族健康人群比较,分析其遗传特点。方法 54例冠心病患者的DNA应用ACE基因内含子16多态部位侧翼的前体进行PCR扩增。等位基因在琼脂胶上电泳,溴乙啶染色,紫外灯下观察。结果患者的DD,DI和Ⅱ基因型的频率是0.57,0.18和0.24、汉族健康人群的DD,DI和Ⅱ基因型的频率是0.33,0.31和0.35。冠心病患者DD基因型及DD等位基因频率显著增高。结论在北方汉族人群的研究中发现ACE基因I/N的多态性与冠心病相关。
Objective To investigate the polymorphism of ACE gene I / N in patients with coronary heart disease in Han nationality in Inner Mongolia, and to analyze the genetic characteristics of ACE gene in Chinese Han healthy people. Methods The DNA of 54 patients with coronary heart disease was amplified by PCR using the flanking precursors of intron 16 polymorphic region of ACE gene. The alleles were electrophoresed on agar, stained with ethidium bromide and observed under UV light. Results The frequencies of DD, DI and Ⅱ genotypes in patients were 0.57, 0.18 and 0.24, respectively. The frequencies of DD, DI and Ⅱ genotypes in healthy Han population were 0.33, 0.31 and 0.35. Patients with coronary heart disease DD genotype and DD allele frequency was significantly higher. Conclusions In the northern Chinese population, it was found that ACE gene I / N polymorphism is associated with coronary heart disease.
