目的探讨胸腺基质细胞(TSC)对胸腺细胞的调节作用。方法应用光镜、电镜和流式细胞术等,观察初代培养的TSC对胸腺细胞发育的调节作用。结果光镜和电镜均可见,TSC可大量粘附和吞噬胸腺细胞,其中部分胸腺细胞发生凋亡。尽管与TSC共培养的胸腺细胞存活率高,凋亡率低,但其总数却显著减少。结论TSC可通过粘附和吞噬作用,清除部分胸腺细胞。
Objective To investigate the regulatory effect of thymic stromal cells (TSC) on thymocytes. Methods The light microscope, electron microscope and flow cytometry were used to observe the regulatory effect of primary culture TSC on thymocyte development. Results Both light microscopy and electron microscopy showed that TSC could adhere to and swallow thymocytes in large quantities, some of which thymocytes were apoptotic. Although the thymocytes co-cultured with TSC had high viability and low apoptotic rate, the total number of thymocytes was significantly reduced. Conclusion TSC can remove some thymocytes through adhesion and phagocytosis.
