在成熟神经系统中 ,嗅觉上皮 (OE)很特殊 ,它能不断产生新的神经元。本文用细胞培养、组织化学和免疫细胞化学技术对成年小鼠的OE进行了研究。实验显示 :双极细胞NF、NSE、MAP2、OMP和tau蛋白免疫染色为阳性 ,但keratin免疫染色为阴性 ,说明双极细胞是神经元。用不同浓度血清的培养基 ,离体培养成年小鼠的OE ,观察碱性成纤维细胞生长因子 (bFGF)和脑源神经营养因子 (BDNF)对OE细胞数量和突起发育的影响。结果显示 :将bFGF与BDNF的组合分别加到 10 %和 5 %血清的OE培养基 (OEM)中 ,增加了双极细胞的数量和比例 ,刺激了双极细胞突起的发育 ,减少了大细胞和梭形细胞。这种组合的效果比bFGF或BDNF单独作用好。在 10 %血清的OEM中 ,神经营养因子的效果比在 5 %血清的OEM中好。
In the mature nervous system, olfactory epithelium (OE) is very special, it can continue to produce new neurons. In this paper, we studied the OE of adult mice using cell culture, histochemistry and immunocytochemistry. Experiments show that: bipolar cell NF, NSE, MAP2, OMP and tau protein immunostaining positive, but keratin immunostaining was negative, indicating bipolar cells are neurons. OE in adult mice was cultured in vitro with different concentrations of serum, and the effects of basic fibroblast growth factor (bFGF) and brain derived neurotrophic factor (BDNF) on the number of OE cells and the development of the neurons were observed. The results showed that the combination of bFGF and BDNF were added to 10% and 5% serum respectively in OE medium (OEM), which increased the number and proportion of bipolar cells, stimulated the development of bipolar cell protrusions and decreased the number of large cells And spindle cells. The effect of this combination is better than bFGF or BDNF alone. In 10% serum OEMs, neurotrophins performed better than OEMs at 5% serum.
