目的:观察碱性成纤维生长因子(bFGF)在视神经中的表达及在视神经损伤后的反应和作用。方法:健康Wistar大鼠20只,雌雄不拘,按处理方式随机分为3组,视神经钳夹伤8只(A组),假伤对照组8只(B组),正常对照4只(C组)。采用大鼠球后视神经钳夹伤模型,利用免疫组化法和计算机图像分析技术,于术后1,3,7,14d检测视神经bFGF的表达。结果:损伤后胶质细胞排列紊乱,胞体增大。视神经损伤后(1,3,7,14d)bFGF阳性细胞表面积密度(%)分别为2.20±0.18,2.18±0.17,2.29±0.45,2.32±0.37,与假伤组(1.52±0.33,1.51±0.37,1.50±0.42,1.48±0.32)和对照组(1.49±0.29,1.50±0.30,1.49±0.35,1.51±0.29)比较,差异有显著性意义(t=3.037~4.158,3.531~4.182,P<0.05)。结论:视神经损伤提高bFGF在视神经中的表达,对视神经损伤的修复具有积极的作用。
Objective: To observe the expression of basic fibroblast growth factor (bFGF) in the optic nerve and its response after optic nerve injury. Methods: Twenty healthy Wistar rats were randomly divided into 3 groups according to the treatment method, including 8 optic nerve crush injuries (group A), 8 sham injury control group (group B) and 4 normal controls (group C) ). The rat model of posterior optic nerve crush injury was used. The expression of bFGF in the optic nerve was detected by immunohistochemistry and computer image analysis at 1, 3, 7 and 14 days after operation. Results: Glial cells were disordered and the cell bodies increased after injury. The area density (%) of bFGF-positive cells after optic nerve injury were 2.20 ± 0.18, 2.18 ± 0.17, 2.29 ± 0.45 and 2.32 ± 0.37, respectively, which were significantly higher than those in sham-injury group (1.52 ± 0.33,1.51 ± 0.37 , 1.50 ± 0.42 and 1.48 ± 0.32 respectively) and the control group (1.49 ± 0.29,1.50 ± 0.30,1.49 ± 0.35,1.51 ± 0.29), the difference was significant (t = 3.037-4.1588, 3.531-4.182, P <0.05 ). Conclusion: The optic nerve injury increases the expression of bFGF in the optic nerve and plays a positive role in the repair of optic nerve injury.
